I recently did cam, timing replacement at 145k noticed oil leak and after doing dye tracked leak to valve cover(repaired no longer leaking) and a second leak from cam shaft seal.
Is it possible to change the seal without having to remove the cover and cam bearing cap #1?

It can be done without removal of either but you would have to fashion a tool to remove the cam hub from the camshaft and then one to dig out the seal from the #1 cam bearing saddle/cap.
Doing it the other way makes it easier although the bolts should be replaced if taking the rocker shaft and bearing cap off.
They are one time use if they are the original VW bolts.

Used a harmonic balancer remover to take cam pulley hub off and long dental picks to remove bad seal. So far 400 miles and no signs of oil leaking.
